These authors describe experiments that identify a substrate docking site on cyclin A. A hydrophobic patch, characterized by the MRAIL sequence, was mutated at several residues and then tested for its ability to interact with substrate proteins containing RXL motifs. These data suggest that the hydrophobic patch is involved in substrate recruitment of cdk2. The addition of a second substrate docking site, an LXCXE motif, onto the amino-terminus of cyclin A with substitutions in its hydrophobic patch restored substrate targeting.

This paper presents the first crystal structure of a viral cyclin. The solved structure shows that V cyclin has an overall structure that is similar to that of cyclin A and cyclin H. The authors model a V cyclin/cdk2 model based on the solved structure of cyclin A/cdk2. They propose that the lack of binding to CKIs is due to a change in the shape of the substrate binding pocket as well as substitutions of critical contact residues within the cyclin.

Transformation of primary human endothelial cells by Kaposi's sarcoma-associated herpesvirus
-
The authors describe the ability of KSHV to infect and transform primary human enodothelial cells. Infected endothelial cells have a spindle-shaped morphology and continue to survive for months after infection, as a result in part of a continued responsiveness to VEGF. At sub-physiological concentrations of VEGF, infected cells underwent lytic replication producing progeny virions. Remarkably, only 5% of the cells in culture contained KSHV DNA, and the uninfected cells survive via a paracrine effect. Uninfected cells treated with conditioned media from KSHV-infected cells assume a spindle-shaped morphology and upregulate the VEGF receptor. KSHV-infected cells had telomerase activity and were capable of anchorage-independent growth.

The authors describe the ability of KSHV to infect and transform primary human enodothelial cells. Infected endothelial cells have a spindle-shaped morphology and continue to survive for months after infection, as a result in part of a continued responsiveness to VEGF. At sub-physiological concentrations of VEGF, infected cells underwent lytic replication producing progeny virions. Remarkably, only 5% of the cells in culture contained KSHV DNA, and the uninfected cells survive via a paracrine effect. Uninfected cells treated with conditioned media from KSHV-infected cells assume a spindle-shaped morphology and upregulate the VEGF receptor. KSHV-infected cells had telomerase activity and were capable of anchorage-independent growth.

This report describes experiments that test the contribution of G-protein-coupled receptor kinase to KSHV-mediated oncogenesis. The KSHV-GPCR causes focus formation in NIH3T3 cells, which can be inhibited by certain GPCR-specific kinases. Focus-derived cells, when injected into nude mice, can develop into tumors. Conditioned medium supporting the growth of GPCR-transfected cells was positive in a microtubule-formation assay as well as a co-culture matrigel assay. This conditioned media contained higher levels of VEGF, which appears to mediate an angiogenic response.

The effect of cyclin expression on thymus and T cell development, as well as its effects on proliferation and apoptosis, are described. Notably, the expression of viral cyclin correlates with the development of high-grade lymphoblastic lymphoma in 40% of transgenic mice.
