Misshapen and its mammalian homolog, NIK, require the small GTPase Rac to activate the JNK pathway in cell cultures but, unlike other Ste20-like mechanisms, Misshapen does not bind activated Rac directly, suggesting alternative mechanisms.

The authors here identify the first Drosophila TRAFs, DTRAF1 and DTRAF2, and show that DTRAF1, but not DTRAF2, binds and activates Misshapen (see [22•]) in cell cultures. They further show that the mammalian homolog of Misshapen, NIK, also associates with TRAFs, suggesting a conserved link between TRAFs and Ste20 kinases.
