These investigators demonstrate that transgenic mice expressing a mutated PSD-95 have dramatically altered spatial learning and synaptic plasticity, although synaptic structure and NMDA receptor localization appear normal. These observations suggest that the primary role of PSD-95 in vivo may be to connect the NMDA receptor to downstream signaling mechanisms and not, as predicted from heterologous cell assays, the localization or organization of these receptors at synapses.

The authors demonstrate that PSD-95 and other MAGUKS can form homomultimers and heteromultimers via cysteine bonds and that multimerization is required for PSD-95 to bind to two different receptors. These observations suggest a model whereby transmembrane ion channels and receptors might be clustered by multimerization of underlying scaffolding proteins.

One of several recent papers of note describing novel synaptic PDZ proteins that have receptor specificities distinct from those of the MAGUKs. PICK1 is a protein with a single PDZ domain that can bind to the carboxyl terminus of AMPA receptors. It colocalizes with these receptors at excitatory synapses and can cluster them in a heterologous cell assay. PICK1 also binds to protein kinase C and the Eph family of receptor tyrosine kinases. These results and the identification of other novel scaffolding proteins like Homer and GRIP suggest a second scaffold involved in organizing synaptic proteins. How these proteins interact with other scaffolding proteins, like the MAGUKs, is unclear at present.

These receptors are critical for axonal guidance in the developing nervous system. The PDZ proteins colocalize with Eph receptors at synapses, and cluster receptors in a heterologous cell assay, suggesting that PDZ proteins organize Eph receptors and their ligands at synapses in vivo.

Notably, Mint1 had already been characterized as a binding partner of Munc18-1, a protein implicated in synaptic vesicle exocytosis. This raises the possibility that the complex conserved in both C. elegans and vertebrates might regulate targeting of proteins to specific membrane domains.
