Coassembly of TRP and TRPL produces a distinct store-operated conductance
-
of special interest. A combination of biochemical and electrophysiological approaches were used to demonstrate that TRP and TRPL form heteromultimeric channels. The significance of this observation is that association of TRP with TRPL is critical for suppressing the constitutive activity of TRPL and formation of a store-operated channel. The properties of TRP/TRPL are distinct from the TRP store-operated channel. Thus, heteromultimeric assembly among TRP family members provides a mechanism for the production of a multitude of store-operated channels with a diversity of biophysical characteristics.

Furthermore, this work demonstrated that the association of TRP with INAD is required for the rhabdomere-specific localization of TRP in vivo.

Synaptic clustering of the cell adhesion molecule Fasciclin II by Discs-Large and its role in the regulation of presynaptic structure
-
of special interest. FASII binds directly to discs-large protein (DLG) and requires this interaction for proper localization at type I synapses at the Drosophila neuromuscular junction. Mutations in either fasII or dlg result in a similar morphological defect - an increase in the number of active zones - consistent with a role of each locus in the same developmental pathway. Evidence is also provided that DLG, Shaker and fasciclin II form a ternary complex.

Coordination of an array of signaling proteins through homo- and heteromeric interactions between PDZ domains and target proteins

Xu X-Z, Choudhury A, Li X, Montell C. Coordination of an array of signaling proteins through homo- and heteromeric interactions between PDZ domains and target proteins. of special interest J Cell Biol. 1998; Rhodopsin, TRPL, PKC and calmodulin bind directly to INAD. As TRP and PLC have previously been shown to bind calmodulin, a total of at least six proteins functioning in phototransduction are tethered to INAD. In addition, INAD homomultimerizes, leading to the formation of INAD polymers. Thus, the signaling complex that functions in fly vision appears to be a massive web consisting of INAD and most of the components required in phototransduction.

Two distantly positioned PDZ domains mediate multivalent INAD - Phospholipase C interactions essential for G protein-coupled signaling

of special interest. The PLC encoded by the NORPA locus is linked to INAD via two PDZ domains. Binding to PDZ1 occurs through the carboxyl terminus of the PLC whereas association to PDZ5 is mediated by an internal sequence.
