Immunity 1998, 8:255-264. IL-4Rα-deficient and Stat6-deficient mice fail to expel N. brasiliensis whereas IL-4-deficient mice expel this parasite. Both IL-4 and IL-13 contribute to N. brasiliensis expulsion but IL-13 appears to be more important for expulsion than IL-4. Stat6 is not required for IgG1 responses to this parasite and inhibits IL-4 mediated induction of mucosal mastocytosis.

Expression of the transcription factor GATA-3 and the cell surface receptor CD30 are more severely affected in IL-4Rα-deficient mice than in IL-4-deficient mice. Injection of recombinant IL-13 induces worm expulsion in mice in which a defect in the Rag2 gene causes B and T lymphocyte deficiency.

Th2 cells generated using T cells from IL-13-deficient mice make less IL-4, IL-5 and IL-10 than wild-type mice whereas mast cells from these mice make similar levels of type 2 cytokines. In vitro type 2 cytokine responses are not rescued by added IL-13 but in vivo addition of IL-13 can increase Th2 responses, as can N. brasiliensis infection. IL-13 deficient mice have lower basal serum IgE levels and biased antigen specific immunoglobulin isotype responses.

Chiaramonte MG, Schopf LR, Neben TY, Cheever AW, Donaldson DD, Wynn TA: IL-13 is a key regulatory cytokine for Th2 cell-mediated pulmonary granuloma formation and IgE responses induced by Schistosoma mansoni eggs. J Immunol 1999, 162:920-930. In vivo blockade of IL-13 reduces pulmonary granuloma size and total serum IgE levels in mice injected with S. mansoni eggs but did not affect cytokine responses whereas IL-4 deficient mice make lower IL-13 levels than wild-type mice. Granuloma size is also reduced in IL-4 deficient mice and granulomas and pulmonary eosinophilia are nearly absent in IL-13 suppressed, IL-4 deficient mice.

Overexpression of IL-13 on a lung-specific promoter induces an eosinophil and mononuclear pulmonary infiltrate, multinucleated pulmonary macrophages, airway epithelial cell hyperplasia, mucus cell metaplasia, increased mucus production, subepithelial airway fibrosis, increased eotaxin production, increased baseline airway resistance and hyper-responsiveness to methacholine.

Nasal administration of either IL-4 or IL-13 to mice induces an IL-4Rα dependent, asthma like phenotype with airway hyper-responsiveness, eosinophil and neutrophil infiltrates and goblet cell hyperplasia in wild-type mice and mice deficient in B and T cells. Neutralization of endogenously produced IL-13 inhibits airway hyper-responsiveness, pulmonary eosinophilia and goblet cell hyperplasia in allergen primed mice that then inhale the same allergen.
