    • Urban JF Jr, Noben-Trauth N, Donaldson DD, Madden KB, Morris SC, Collins M, Finkelman FD: IL-13, IL-4Rα, and stat6 are required for the expulsion of the gastrointestinal nematode parasite Nippostrongylus brasiliensis. Immunity 1998, 8:255-264. IL-4Rα-deficient and Stat6-deficient mice fail to expel N. brasiliensis whereas IL-4-deficient mice expel this parasite. Both IL-4 and IL-13 contribute to N. brasiliensis expulsion but IL-13 appears to be more important for expulsion than IL-4. Stat6 is not required for IgG1 responses to this parasite and inhibits IL-4 mediated induction of mucosal mastocytosis.

    • Differences between IL-4Rα-deficient and IL-4-deficient mice reveal a role for IL-13 in the regulation of Th2 responses
    • Barner M, Markus M, Brombacher F, Kopf M: Differences between IL-4Rα-deficient and IL-4-deficient mice reveal a role for IL-13 in the regulation of Th2 responses. Curr Biol 1998, 8:669-672. IL-4Rα, unlike IL-4, is required for control of N. brasiliensis infection and Th2 development during infection - as characterized by cytokine production. Expression of the transcription factor GATA-3 and the cell surface receptor CD30 are more severely affected in IL-4Rα-deficient mice than in IL-4-deficient mice. Injection of recombinant IL-13 induces worm expulsion in mice in which a defect in the Rag2 gene causes B and T lymphocyte deficiency.

Both IL-4- and IL-13-deficient mice are susceptible to infection with T. muris whereas wild-type littermates are resistant. IL-4-deficient mice make decreased Th2 responses and failed to produce IgG1 antibodies whereas IL-13-deficient mice first make low, then later make normal, Th2 responses but are unable to regulate the magnitude of their antibody isotype response.

Th2 cells generated using T cells from IL-13-deficient mice make less IL-4, IL-5 and IL-10 than wild-type mice whereas mast cells from these mice make similar levels of type 2 cytokines. In vitro type 2 cytokine responses are not rescued by added IL-13 but in vivo addition of IL-13 can increase Th2 responses, as can N. brasiliensis infection. IL-13 deficient mice have lower basal serum IgE levels and biased antigen specific immunoglobulin isotype responses.

J Immunol 1999, 162:920-930. In vivo blockade of IL-13 reduces pulmonary granuloma size and total serum IgE levels in mice injected with S. mansoni eggs but did not affect cytokine responses whereas IL-4 deficient mice make lower IL-13 levels than wild-type mice. Granuloma size is also reduced in IL-4 deficient mice and granulomas and pulmonary eosinophilia are nearly absent in IL-13 suppressed, IL-4 deficient mice.
