These two references provide experimental evidence that iGluR receptors exist as tetrameric complexes rather than pentameric subunit complexes, as was previously supposed. The findings of these papers are not final proof, and further physical evidence is required to clarify the issue.

The authors describe a crystal structure of the iGluR-binding region in a complex with the neurotoxin kainic acid. This is a seminal paper and confirms much of which was modelled previously using E. coli glutamine-binding protein. The report identifies determinants of receptor-agonist interactions and describes how ligand specificity and affinity can be affected by remote amino acid residues. Understanding the recognition site at this level is critical for drug discovery efforts.

The discovery of phenylsulfonyl-aminoethylthio-2,6-difluoro-phenotyncetamide (PEPA) as an allosteric potentiator of AMPA receptors with the flop splice variant is useful to dissect receptor heterogeneity. PEPA is complementary to the action of cyclothiazide that prefers flip splice variants, and is potentially useful for investigating new therapeutic agents as AMPA receptors with the flop variant are found more frequently in mature receptor populations.
