This study shows that targeted expression of a carboxy-terminal peptide of the alpha subunit of Gq to the heart reduces cardiac hypertrophy in response to pressure overload. The study demonstrates the importance of Gq signaling in pressure overload hypertrophy and suggests that inhibitors of Gq may be useful in the treatment of heart failure.

Expression of a β-ARK inhibitor in the heart of transgenic mice is here shown to restore new cardiac function and prevent chamber enlargement in response to null mutation of the muscle LIM domain protein (MLP). These results suggest an important role for β-adrenergic receptor/G protein coupling in heart failure and suggest possible therapeutic approaches involving β-ARK inhibition.

The authors of this paper demonstrate that activated calcineurin and activated NFAT3 are sufficient to activate cardiac hypertrophy in vivo. The results show that the calcineurin inhibitor, cyclosporin A, blocks Ang II and PE-mediated cardiac hypertrophy in vitro. These studies uncover a novel hypertrophic signaling pathway connecting the membrane to the nucleus.

This paper demonstrates that calcineurin activation and the resulting dephosphorylation of NFAT occurs specifically in response to a low, sustained calcium plateau, whereas activation of NF-κB by c-Jun amino-terminal Kinase (JNK) occurs in response to a large transient calcium rise. These results demonstrate how intracellular calcium-regulated signaling systems can distinguish between different types of calcium signals and thereby activate specific transcriptional programs in the nucleus. The results are especially relevant to understanding the role of calcium in cardiac hypertrophy.

The authors report that cardiomyopathy in response to misexpression of sarcomeric proteins in the heart can be prevented by systemic cyclosporin A administration. These results suggest that at least certain forms of hypertrophy arising from altered sarcomeric functions may involve calcineurin activation. The study also demonstrates that pressure overload hypertrophy in rats can be prevented by cyclosporin A, further implicating calcineurin in this process.
