Clinical outcome in diffuse large B-cell lymphoma is dependent on the relationship between different cell-cycle regulator proteins
-
Sanchez E, Chacon I, Plaza MM, Munoz E, Cruz MA, Martinez B, et al.: Clinical outcome in diffuse large B-cell lymphoma is dependent on the relationship between different cell-cycle regulator proteins. J Clin Oncol 1998, 16:1931-1939. Demonstrates the adverse effect of bcl-2 expression on outcomes of a clearly defined patient population with diffuse large cell lymphoma treated with doxorubicin-based therapy. The detection of t(14;18) does not affect outcome and is less frequent than bcl-2 expression. This suggests that bcl-2 is upregulated in many patients by mechanisms other than the t(14;18).

Confirms the association of bcl-2 expression with inferior outcome in aggressive lymphomas. References 22 (Sanchez et al: J Clin Oncol 1998, 16:1931-1939.) and 23, coupled with in vitro work (Sundaresan et al.: Cell Growth Differ 1997, 8:1071-1082) suggest that pharmacologic down regulation of bcl-2 levels by retinoids may be therapeutically useful.

Br J Heamatol 1999, Haematologica 1999, 84:116. Elevation of serum IL-10 is associated with inferior failure-free survival in Hodgkin's disease. Similar results have been reported for non-Hodgkin's lymphomas. Elucidation of the involved mechanisms may provide targets for therapeutic intervention. These may include monoclonal antibodies against IL-10 or its receptor, IL-10 receptor-binding antagonists, or pharmacologic down-regulation of bcl-2 levels.

Blood 1998, 91:3011-3016. Soluble CD30 may be useful both as a prognostic feature and as a tumor marker in Hodgkin's disease. It has already been shown to be a tumor marker and prognostic in patients with ALCL.
