16. 7 cells, LeukoSite (3)] were pelleted, resuspended in 500 μl of envelope glycoprotein-containing supernatants, and rocked gently at 37°C for 1 hour. Cells were pelleted, washed twice in PBS, and lysed by the addition of NP-40 buffer (0.5 M NaCl, 10 mM tris, pH 7.5, 0.5% NP-40). Lysates were cleared (20,000g at 4°C for 15 min) in a microcentrifuge, and the envelope glycoproteins were precipitated overnight at 4°C by a mixture of sera from HIV-1-infected individuals and protein A-Sepharose. Sepharose pellets were washed in NP-40 buffer, boiled in SDS-containing sample buffer, and run on 12.5% SDS-polyacrylamide gels. Autoradiographed gels were quantitated with a densitometer.
