This paper describes the use of retroviral vectors to transduce primary myoblasts with reverse tetracycline transactivator (rtTA) and inducible erythropoietin (Epo). Following implantation into C3H mice, Epo secretion could be switched on and off in a doxycycline-dependent manner. There was no apparent immune response against rtTA expression recipient animals, as Epo secretion persisted for five months.

This publication describes the development and evaluation of one- and two-retrovirus based systems for transduction of tetracycline-controlled transactivator or reverse tetracycline transactivator and an inducible gene of interest. Similar to experiments described in [11], doxycycline-dependent induction of human growth hormone secretion could be observed at 16 and 46 days after transplantation into recipient mice.

Significant dose-dependent induction of human growth hormone secretion was noted when mice received the progesterone antagonist RU486 orally or through intraperitoneal injection. The use of chromosomal insulator sequences to minimize position effects on GLVP transactivator expression is also described.

A mammalian three-hybrid screen was performed to identify compensatory mutations in the rapamycin-binding domain of FRAP to restore binding to these modified derivatives. Inclusion of these components into dimerization-based strategies of gene regulation may increase specificity and decrease toxicity currently associated with the system.
