This paper describes the development of a gene switch based on a chimeric Drosophila EcR. For maximum activity, the chimeric receptor must be coexpressed with a member of the RXR family. Inductions as high as 20,000-fold were reported. Tissue-specific expression of the gene switch and drug-dependent target gene regulation were also demonstrated in transgenic mice.

This paper describes a number of significant improvements in the antiprogestin-activated gene switch as well as a ligand-dependent transcriptional repressor. Modifications in the length of the LBD and the position of the major transactivation domain generated a significantly more potent ligand-dependent transcriptional activator and a 10-fold reduction in threshold concentration for the ligand.
