    • Major role of the histones H3-H4 in the folding of the chromatin fiber
    • of special interest. Using a reassembled defined polynucleosome array lacking histone H1, this paper and [4] show the complexity of tail domain function in chromatin fiber folding. Both studies provide evidence that the H3 and H4 tails are dominant mediators of chromatin fiber folding; however, [4] shows that extensive folding of the fiber requires the tail domains of the four core histones.

    • Histone H1b phosphorylation is dependent upon ongoing transcription and replication in normal and ras-transformed mouse fibroblasts

This and an earlier study show that phosphorylation of a mouse histone H1 subtype (H1b) is elevated in oncogene-transformed cells, most likely through activation of the mitogen-activated protein kinase pathway. Phosphorylated H1b colocalized with centres of RNA splicing. It is demonstrated that phosphorylation of H1b is dependent upon ongoing transcription and replication. No other histone modification is known to be dependent upon these two processes.

These studies suggest that a ligand-activated nuclear receptor recruits several classes HATs to counteract the repressive action of the histone deacetylase recruited by steroid receptor corepressors (N-CoR, SMRT).

AIB1 is frequently amplified and overexpressed in estrogen receptor positive breast cancer cell lines and in breast tumors. The sequence of AIB1 is identical to ACTR (see [19]). Thus, AIB1 is a HAT that recruits P/CAF and CBP and is highly expressed in breast cancer cells. Together, these studies [19,25] contribute to the emerging role for chromatin modulators in oncogenesis (see also [47,48]).

Furthermore, [45-47] show that mSin3A binds to the corepressors N-CoR and SMRT. In yeast, the Sin3-RPD3 complex is recruited by the DNA-binding protein Ume6 which regulates genes involved in meiosis [50]. In mammalian cells, Mad-Max heterodimers, which bind to E-box-related DNA sequences, and nonliganded nuclear receptors (retinoic acid and thyroid hormone receptors) recruit the large N-CoR (SMRT)-mSin3A-HDAC1(2) complex, demonstrating a convergence of repression pathways for nonliganded nuclear receptors and basic region helix-loop-helix leucine zipper (bHLH-Zip; e.g. Mad-Max) proteins [41-47]. In [41], the authors show that mSin3A (150 kDa) is in a stable, high molecular mass, multiprotein complex. In addition to HDAC1(2), the complex(es) most likely contain N-CoR (270 kDa), SAP30, SAP18, and RbAp46,48 [44,46].
