This paper provides the first crystal structure of a nuclear receptor ligand-binding domain. The overall structure has proven to be prototypic for the ligand-binding domains of other nuclear receptors. As the only available structure for an unliganded LBD, comparison with the structures of the liganded RAR and liganded TR LBDs suggests the nature of the conformational changes in receptor structure that are induced by the binding of ligand.

This paper describes the crystal structure of the liganded RAR LBD. The overall fold of the LBD is shown to be similar to that of the RXR LBD. The structure illustrates the relationship between all-trans retinoic acid and the ligand-binding pocket. The AF-2 region of the LBD, which is required for transcriptional activation, makes direct contact with the ligand, suggesting that it may play a structural role, in addition to a potential role in interacting with coactivator molecules. By comparing the liganded RAR structure with the unliganded RXR structure [6••], the authors suggest the nature of the conformational changes that might be induced in the LBD by retinoic acid.

As in [7••], the overall fold of the LBD is shown to be similar to that of the RXR LBD, but is much more compact, suggesting a structural role of the ligand. As in the structure of the liganded RAR, the AF-2 region of the liganded TR is shown to be tightly packed against the ligand-binding domain and to contribute to the ligand-binding pocket.

In this paper, the authors demonstrate that p300 associates with the estrogen receptor in a ligand-dependent and AF-2-dependent manner. The association of p300 with the estrogen receptor is induced by estradiol, but not by an estrogen receptor antagonist. p300 is also shown to potentiate estrogen-dependent transcription, suggesting that it is a component of a nuclear receptor coactivator complex.

The authors of this paper demonstrate direct interactions between CBP/p300 and several nuclear receptors, in addition to direct interaction between CBP/p300 and SRC-1/N-CoA1 CBP/p300 are demonstrated to play essential coactivator roles for the RAR, on the basis of inhibition of RA-dependent transcription by microinjection of anti-CBP antibodies. Over-expression of CBP is demonstrated to relieve inhibition of AP-1 activity by retinotc acid, suggesting that AP-1 antagonism reflects competition for limiting amounts of CBP/p300 in cells. A full-length cDNA encoding SRC-1/N-CoA1 reveals that the amino terminus contains a PAS homology domain and represents a component of biochemically defined p160.

TIF2 potentiates ligand-dependent transcription by several nuclear receptors and is shown to contain an intrinsic activation domain.
