Two forms of two-dimensional RNA polymerase II crystals were obtained. The enzyme lacking subunits 4 and 7 represents the open form allowing entry of DNA whereas the complete enzyme shows the closed state which may represent the form during transcription elongation.

The underproduction of one subunit of RNA polymerase II was found to influence cell growth, indicating that the synthesis of RNA polymerase II subunits is coordinated and that the concentration of RNA polymerase is maintained at a minimum level.

Nucleic Acids Res 1996, 24:4543-4551. The promoter-upstream regions of the RPB1 and RPB2 genes of the budding yeast carry the binding site sequences for transcription activators, Rep1 and Rep2. This finding suggests that the regulation of RNA polymerase synthesis is growth-related, as in the case of ribosomes.

This paper together with Fan et al., 1996 [41•] describes evidence for direct involvement of the recombination components in the formation of RNA polymerase holoenzyme. Linking between recombination and transcription can be explained by this finding.

After analysis of RNA polymerase II complexes isolated using anti-carboxy-terminal domain antibodies, the authors found a novel RNA polymerase II complex, different from the known holoenzyme, which may be involved in transcription of a different set of genes.
