In response to angiotensin II, PKC-z associates with Ras in vascular smooth muscle cells. Antisense PKC-z oligonucleotide treatment resulted in depletion of PKC-z protein and concentration-dependent decreases in angiotensin II-dependent ERK1/2 activation. By contrast, inhibition of PKC-z expression had no effect on phorbol ester or platelet derived growth factor-induced activation of ERK1/2.

Thus oligomerization of Raf promotes Raf activation through a Ras-dependent mechanism.

The authors find an increase in glomerular MEK and ERK activities over prolonged periods of time in an accelerated model of anti-glomerular basement membrane glomerulonephritis in rats. In contrast, stress-activated protein kinase activities were activated only in the early stages of the disease.

This pathway of activation, together with MEKK1 dependent activation of NFκ-B, act together to result in maximal expression of E-selectin.
