The DNA-binding domain of Gal4 (amino acids 3 through 147) was amplified by polymerase chain reaction and ligated in frame with wild-type or mutant murine CHOP complementary DNA (cDNA) at the unique Nhe I site of the mutant cDNA, thus deleting the CHOP dimerization domain. The chimeric cDNA was transferred to the pCDNA-Amp mammalian expression plasmid.

Lipid content was estimated by measuring the integrated intensity of the red signal in scanned images of the plates and is presented in arbitrary units (Fig. 3D). Shown is a typical result of an experiment performed in duplicate and reproduced three times. Equal expression of the wild-type and mutant proteins was observed by immunocytochemistry and immunoblotting (X.-Z. Wang, unpublished data).
