Although t-PA gene expression subsided within 7 days, it effectively increased plasma fibrinolytic activity-restoring thrombolytic capacity in t-PA-deficient mice and augmenting thrombolytic capacity in PAI-1-overexpressing mice.

This study describes an attempt to redirect infarcted myocardium to form skeletal muscle instead of scar tissue by injecting an Ad5 vector carrying a myogenic determination gene (MyoD) into cardiac granulation tissue. When reparative tissue was treated at high dose, the MyoD gene induced the expression of other muscle-specific genes (myogenin and embryonic skeletal MHC), suggesting a strategy that might eventually provide new contractile tissue to repair myocardial infarcts.

This study examines the effect of injecting an Ad5 vector expressing human preproendothelin-1 (preproET-1) into the caudal vein of rats. After 4 days, hepatic overexpression of preproET-1 caused a sixfold elevation in circulating plasma ET-1 levels and a 28 mm Hg increase in mean arterial blood pressure. Intravenous infusion of an ET(A) receptor antagonist reduced blood pressure in these rats, indicating that endogenous overexpression of preproET-1 can cause systemic hypertension through the activation of the ET(A) receptor.
