This study describes an attempt to redirect infarcted myocardium to form skeletal muscle instead of scar tissue by injecting an Ad5 vector carrying a myogenic determination gene (MyoD) into cardiac granulation tissue. When reparative tissue was treated at high dose, the MyoD gene induced the expression of other muscle-specific genes (myogenin and embryonic skeletal MHC), suggesting a strategy that might eventually provide new contractile tissue to repair myocardial infarcts.

This study examines the effect of injecting an Ad5 vector expressing human preproendothelin-1 (preproET-1) into the caudal vein of rats. After 4 days, hepatic overexpression of preproET-1 caused a sixfold elevation in circulating plasma ET-1 levels and a 28 mm Hg increase in mean arterial blood pressure. Intravenous infusion of an ET(A) receptor antagonist reduced blood pressure in these rats, indicating that endogenous overexpression of preproET-1 can cause systemic hypertension through the activation of the ET(A) receptor.

This study used an Ad5 vector to overexpress the sarcoplasmic reticulum calcium ATPase (SERCA2a) in neonatal rat myocytes and found alterations in intracellular calcium handling that resulted in enhanced contraction as measured by cell shortening measurements.
