    • Structural insights into the evolution of an antibody combining site
    • ••], showing by X-ray crystallography the structural differences of the germline and affinity-matured antibodies both with and without hapten. It indicates that conformational changes of the germline antibody are required in order to bind the hapten, however, no such changes are observed when the hapten binds to the affinity-matured antibody. This suggests that upon affinity maturation, the cavity becomes preorganised to encourage stronger binding of the hapten.

    • Janda KD, Lo L-C, Lo C-HL, Sim M-M, Wang R, Wong C-H, Lerner RA: Chemical selection for catalysis in combinatorial antibody libraries. Science 1997, 272:945-948. The authors demonstrate a method of direct chemical selection for catalysis from a phage-antibody library. This is achieved by releasing a reactive product, which is attached to a support, after the reaction has been catalysed, allowing the phage carrying catalytic antibody to be 'trapped'. The 'trapped' catalytic antibody can then be released and isolated.

    • Hollfelder F, Kirby AJ, Tawfik DS: Off-the-shelf proteins that rival tailor-made antibodies as catalysts. Nature 1997, 383:60-63. The authors demonstrate that in at least one case - and by implication, more generally - a reaction can be catalysed just as well by off-the-shelf proteins as by selected catalytic antibodies. This paper suggests that precise positioning of the catalytic site is less important than cavity medium effects.
