RAIDD is a new 'death' adaptor molecule
-
of special interest. The set of papers [42-45,46-49] describe the isolation of a series of signal transducers for Fas- or TNFR1-mediated apoptosis and demonstrate protein-protein interactions via the death-domain bridge Fas or TNFR1 to the inactive form of the ICE-like proteases. The overlaps and specificities of Fas or TNFR1 which mediate apoptosis signals appear to be determined by which signal adapter proteins are recruited to the receptor.

TNF-α And cancer therapy-induced apoptosis: Potentiation by inhibition of NF-κB
-
of outstanding interest. This paper, along with [57-59], shows that activation of NF-κB is able to work as an anti-apoptotic signal for TNF-induced cell death, suggesting a molecular mechanism for the prevention of apoptosis during inflammation. The results also present a mechanism explaining why TNF-induced apoptosis can be accomplished in many cell types only in the presence of cycloheximide.
