This paper describes the in vivo packaging of retrovirus particles in baby hamster kidney (BHK) cells with the aid of the Semliki Forest virus (SFV) replicon. RNA from retroviral genes on separate SFV vectors transfected into BHK cells resulted in high titer retroviral yields, showing protease and reverse transcriptase activities. This technology opens up new possibilities for using retroviruses in gene therapy.

In this study, the familial Alzheimer's disease related gene presenilin 1 was expressed in BHK cells and NT2N primary neurons by infection with recombinant Semliki Forest virus particles. Immunoreactive presenilin 1 was detected in cell bodies and dendrites of the neurons and could be colocalized with the rough endoplasmic reticulum marker protein calnexin in BHK cells.

J Virol 1996, 70:1953-1960. This paper describes the immunization of pigtail macaques with either recombinant SFV expressing SIV-PBj14Env gp160 or purified recombinant SIV-PBj14 gp120. The results indicated that the immunizations did not protect against virus infection but lowered the virus load in plasma and protected against the lethal challenge with SIV-PBj14 virus.

This paper describes the application of Venezuelan equine encephalitis virus (VEE) as a vaccine vector. Efficient immune responses were obtained, because VEE replicates primarily in lymphoid tissue. Inoculation of VEE virus expressing the influenza virus hemagglutinin gene induced both anti-hemagglutinin IgG and IgA antibodies. The vaccinated animals showed complete protection when intranasally challenged with a virulent influenza virus strain.

In this study the recombinant protein production is carried out at one liter scale using Semliki Forest virus vectors. Reproducible high level expression of enzymatically active human cyclo-oxygenase 2 was achieved. The conditions for efficient infection of BHK cells in suspension were optimized. Important factors were the pH and serum concentration of the medium and the multiplicity of infection.

A single point mutation (Pro726→Ser) in the nonstructural P2 (nsP2) gene was able to establish persistent infection in BHK cells. Introduction of the nsP2 mutation in the Sindbis virus vector resulted in higher levels of β-galactosidase expression.
