These cells were maintained in RPMI 1640 culture medium containing 10% FBS and antibiotics (100 U/mL penicillin G and 100 μg/mL streptomycin). All experiments were carried out 24 h after cells were seeded. Chemical compounds were dissolved in DMSO and diluted with DMEM medium to final concentrations of 1 nM to 100 μM. OVCAR-8 and NCI/ADR-RES cells were incubated with chemical compounds for 48 h and then the MTT assay was performed. A 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyl tetrazolium bromide (MTT) reduction was determined to examine the effect of chemical compounds on cell viability. Briefly, following the treatment of cells with different concentrations of chemical compound (final concentration, 1 nM, 10 nM, 100 nM, 1 μM, 10 μM, and 100 μM) for 48 h, cells were incubated with MTT solution (0.5 mg/mL) for 3 h at 37 oC. The violet precipitate (formazan) in MTTtreated cells was dissolved with 100 μL of DMSO and then analyzed at a wavelength of 595 nm with a microplate reader (Beckman Coulter, Brea, CA). Each assay was performed in triplicate.
