    • Significant losses of material due to protein precipitation were observed under the copper-activated click reaction conditions using the alkyne reagents. Similar observations have been reported in the references below. To overcome these problems, we conducted the first click reaction of the PAL reaction products of 1a/2b or 1b/2a with a clyclooctyne reagent under the copper-free condition following the PAL step and the second click reaction with an azide reagent under the copper-activated condition in-gel after SDS-PAGE (in-gel click reaction)
