BRN-103, a novel nicotinamide derivative, inhibits VEGF-induced angiogenesis and proliferation in human umbilical vein endothelial cells

    • Cellular proliferation was determined by BrdU incorporation assay using a commercial ELISA ki according to the manufacturer's instructions (Roche, Basel, Switzerland).     • Mouse aortas isolated from Sprague-Dawley rats (NIH) were cleaned of periadventitial fat and cut into rings at 1-1.5 mm in circumference. The aortic rings were randomized into Matrigel-coated wells and sealed with a 100 μl overlay of Matrigel. VEGF, with or without indicated concentrations of BRN-103 or Sunitinib, was added to the wells. As a control, medium alone was assayed. After 7 day, we fixed the microvessel sprouting and photographed it using an Olympus inverted microscope (Olympus; magnification, × 100).
