Prevention of transthyretin amyloid disease by changing protein misfolding energetics
-
A comparison of pharmacologic and genetic evidence demonstrating that kinetic stabilization of TTR should prevent amyloid disease. This manuscript describes a series of transthyretin amyloidosis inhibitors that functioned by increasing the kinetic barrier for dissociation. The trans-suppressor mutation, T119 M, which is known to ameliorate familial amyloid disease, was also shown to function through kinetic stabilization.
