The transposon was successfully removed by subsequent transient expression of PB transposase in the iPSCs without any trace in the integration site. By using a doxycycline (dox)-inducible promoter, the authors also demonstrated that MEFs derived from PB-iPSCs could be reprogrammed very efficiently through the addition of dox.

Efficient and rapid generation of induced pluripotent stem cells from human keratinocytes. Nat Biotechnol 26 (2008) 1276-1284. The authors showed that reprogramming of human keratinocytes is ∼100-fold more efficient and 2-fold faster as compared to human fibroblasts. The keratinocyte-derived iPSCs also had fewer retroviral integrations.
