Two hundred and fifty grams of peeled and sliced potatoes were washed in tap water and boiled in 500 mL distilled water for 30 min or till it could be crushed by a glass rod and the extract was filtered into a beaker (1000 mL) through a muslin cloth. Twenty grams of dextrose was added to the extract and its volume was made up to 1000 mL by adding additional distilled water to make potato dextrose broth. Agar was dissolved to the potato dextrose broth. The medium thus prepared was dispensed into conical flasks and sterilized in an autoclave at 15l bs psi pressure for 20 min after wrapping the cotton plugs with paper. This sterilized PDA medium was stored for further experiment.
