Pitx2 itself is a mediator of Wnt/β-catenin-induced mRNA stabilization by associating with coregulatory complex.

This mutation does not affect the DNA-binding ability of Hesx1 but results in an impaired ability to recruit corepressor Tle1, suggesting that Hesx1-mediated repression is critical for pituitary development.

    • J. Lopez-Rios, K. Tessmar, F. Loosli, J. Wittbrodt, and P. Bovolenta Six3 and Six6 activity is modulated by members of the groucho family Development 130 2003 185 195 Using a two-hybrid screen, the authors identified Tle1 and Aes as interacting proteins of Six3 and Six6. Tle1 and Aes exhibit overlapping expression pattern with Six3 and Six6. Functional studies showed that Tle1 synergizes with Six3/6 and Aes antagonizes Six3/6 in inducing ectopic eye filed. Mutant Six proteins that are unable to interact with Tle1/Aes are defective in initiating ectopic retina formation, suggesting that Tle1/Are are important partners of Six3/6 function.

Geminin, which is a cell-cycle inhibitor by binding to the licensing factor Cdt1, was identified as a direct interacting protein of Six3. Gain- and loss-of-function studies in medaka embryos demonstrate that Six3 and geminin act antagonistically in controlling proliferation and differentiation.

Additional evidence showed that geminin associated with the polycomb complex on the regulatory regions of Hox gene and represses Hox gene expression. Interaction of geminin with Hox proteins inhibits the binding of the Hox proteins to their targets DNA sequences and displaces Cdt1 from geminin-Cdt1 complex.

    • Eya protein phosphatase activity regulates Six1-Dach-Eya transcriptional effects in mammalian organogenesis
    • X. Li, K.A. Oghi, J. Zhang, A. Krones, K.T. Bush, C.K. Glass, S.K. Nigam, A.K. Aggarwal, R. Maas, and D.W. Rose Eya protein phosphatase activity regulates Six1-Dach-Eya transcriptional effects in mammalian organogenesis Nature 426 2003 247 254 This paper demonstrates that Eya protein has an intrinsic protein phosphatase activity that is essential to switch Six1/Dac from transcription repressors to activators. Six1/Dac/Eya directly regulate c-Myc and Gdnf expression and is required for precursor cells proliferation in multiple organs during development.

Six1 induced cell proliferation is dependent on the presence of cyclin A1, providing a tissue-specific mechanism of Six1 action.

    • S.J. Silver, E.L. Davies, L. Doyon, and I. Rebay Functional dissection of eyes absent reveals new modes of regulation within the retinal determination gene network Mol Cell Biol 23 2003 5989 5999 Extensive structure-function analysis of the Drosophila Eya protein was carried out in this study. The transcriptional activation domain was mapped to a proline-, serine-, and threonine-rich (P/S/T-rich) region of Eya protein. Eya protein is capable of homotypic interaction via its N terminus. In addition, it showed that activation of the Ras signaling can potentiate Eya transactivation and that Groucho is a potent repressor of So-Eya-mediated transcription.
