Helin E2F7, a novel E2F featuring DP-independent repression of a subset of E2F-regulated genes EMBO J 22 2003 6289 6298 The authors of this study provide a thorough characterization of E2F7, a new gene member of the E2F family of transcription factors. The most compelling evidence that it acts in the E2F pathway is provided by chromatin immunoprecipitation assays showing that the endogenous E2F7 protein is recruited to a subset of well-characterized E2F targets in a cell-cycle-specific manner.

It also underscores the importance of target gene specificity in differentiating between E2F family members.

    • PRB contains an E2F1-specific binding domain that allows E2F1-induced apoptosis to be regulated separately from other E2F activities
    • F.A. Dick, and N. Dyson pRB contains an E2F1-specific binding domain that allows E2F1-induced apoptosis to be regulated separately from other E2F activities Mol Cell 12 2003 639 649 A biochemical approach is taken to show that pRB and E2F1 interact in a manner distinct from the more general interaction of pRB with E2F1-4 characterized previously. This interaction is specifically disrupted upon DNA damage and agrees with suggestions that control of apoptosis by E2F is specific to E2F1. The results of this study, on the basis of in vitro evidence, await further confirmation in vivo.

Dynlacht E2F integrates cell cycle progression with DNA repair, replication, and G(2)/M checkpoints Genes Dev 16 2002 245 256 We describe here our identification of many new target genes of E2F1 and E2F4, using genome-wide location analysis with a microarray representing the proximal promoter of 1200 human genes. These results indicate that E2F plays a heretofore unrecognized role in the regulation of genes required for DNA repair, chromatin assembly, condensation, and segregation, as well as multiple checkpoints that ensure genomic integrity. They also confirm that the sets of E2F1 and E2F4 target genes significantly overlap.
