    • Cold shock and regulation of surface protein trafficking convey sensitization to inducers of stage differentiation in Trypanosoma brucei
    • Cold shock (≤20 °C) induces expression of procyclins in bloodstream forms and renders stumpy forms responsive to the differentiation triggers, citrate or cis-aconitate at micromolar concentrations, rather than the millimolar concentrations required at 27 °C. A useful lesson that 'standard' conditions may not tell the whole story.

    • Mass spectrometry was used to identify procyclins expressed by tsetse-derived trypanosomes, confirming that GPEET was the predominant procyclin in early procyclic forms and demonstrating that late procyclic forms express only a subset of EP isoforms. It was also shown that the N-termini of all procyclins were proteolytically processed in tsetse, while the dipeptide and pentapeptide repeats remained intact.
