    • Note that there is a correction factor for the relative populations of the two FRET states due to different detection efficiencies on TCCD for different FRET values. Higher FRET values give rise to lower detection efficiencies. Using model DNA samples with FRET efficiencies of 0.8 and 0.5 we have determined a correction factor of 1.88 for the population of the highFRET species respect to the lowFRET. The details of this study will be published elsewhere. From the fit of YFP-Alx647 histograms, the average lowFRET fraction was 0.4, and is then converted into 0.26 by applying this correction factor
