Human intervertebral disc cell morphology and cytoskeletal composition: A preliminary study of regional variations in health and disease
-
Johnson WE, Roberts S: Human intervertebral disc cell morphology and cytoskeletal composition: a preliminary study of regional variations in health and disease. J Anat 2003, 203:605-612. This study documents the existence of F-actin and vimentin containing cell processes in cells derived from patients with discogenic back pain and scoliosis. A pattern towards increased cell processes in scoliotic cells, but not those from painful discs, was reported. The mechanical and biological function of these processes remains unknown.

Bruehlmann SB, Hulme PA, Duncan NA: In situ intercellular mechanics of the bovine outer annulus fibrosus subjected to biaxial strains. J Biomech 2004, 37:223-231. This study provides the first documentation of cell movements within the extracellular matrix of the intervertebral disc. Results indicate that cell movements may be far more complex than originally thought, with evidence of cell translations, rotations, and movements of cell processes relative to the cell body.

Static compression induces zonal-specific changes in gene expression for extracellular matrix and cytoskeletal proteins in intervertebral disc cells in vitro
-
Chen J, Yan W, Setton LA: Static compression induces zonal-specific changes in gene expression for extracellular matrix and cytoskeletal proteins in intervertebral disc cells in vitro. Matrix Biol 2004, 22:573-583. An early study of isolated cell responses to compression in vitro and at the gene expression level, which demonstrate some patterns that are similar and some that are not, to those reported for posttranslational biosynthesis in explants. The observed dissimilarities likely reflect differences in their mechanical, biochemical, and physicochemical environments.

Biological response of the intervertebral disc to dynamic loading
-
Walsh AJ, Lotz JC: Biological response of the intervertebral disc to dynamic loading. J Biomech 2004, 37:329-337. This study is important for revealing that cell responses to dynamic loading, particularly cell death and proteoglycan content, are dependent on both the frequency and magnitude of the applied stress. The lower frequency and higher peak stress magnitudes induced the greatest increases in proteoglycan content and cell death that may relate to the higher deformations that occur under these conditions.

-
Rannou F, Lee TS, Zhou RH, et al.: Intervertebral disc degeneration: the role of the mitochondrial pathway in annulus fibrosus cell apoptosis induced by overload. Am J Pathol 2004, 164:915-924. The authors show that the mechanical overload-induced IVD degeneration is mediated through the mitochondrial apoptotic pathway in IVD cells.

Comparison of neuropathic pain induced by the application of normal and mechanically compressed nucleus pulposus to lumbar nerve roots in the rat
-

J Orthop Res 2003, 21:535-539. This study, and that from this group of investigators, is important for the links between compression load and pain that are revealed. When the mechanically compressed nucleus pulposus tissue was transplanted to the rat nerve roots, the animals experienced a decreased sensitization to pain, providing some of the first evidence that mechanical stimuli have the potential to regulate the perception of pain.

Cyclic tensile stretch modulates proteoglycan production by intervertebral disc annulus fibrosus cells through production of nitrite oxide
-
Rannou F, Richette P, Benallaoua M, et al.: Cyclic tensile stretch modulates proteoglycan production by intervertebral disc annulus fibrosus cells through production of nitrite oxide. J Cell Biochem 2003, 90:148-157. This study is one of the first to study a mechanism for the inhibition of proteoglycan production in IVD cells by tensile stretch. Using a set of nitric oxide synthase inhibitors, the authors conclude that a specific form of nitric oxide synthase, cNOS, may be involved in the downregulation of proteoglycan synthesis, and upregulation of nitric oxide, following tensile stretch.

Annulus cells release ATP in response to vibratory loading in vitro
-
Yamazaki S, Weinhold PS, Graff RD, et al.: Annulus cells release ATP in response to vibratory loading in vitro. J Cell Biochem 2003, 90:812-818. This study is important for demonstrating that ATP release occurs as a signaling mechanism in intervertebral disc cells, and in response to the mechanical stimulation of vibration.
