    • Johnson WE, Roberts S: Human intervertebral disc cell morphology and cytoskeletal composition: a preliminary study of regional variations in health and disease. J Anat 2003, 203:605-612. This study documents the existence of F-actin and vimentin containing cell processes in cells derived from patients with discogenic back pain and scoliosis. A pattern towards increased cell processes in scoliotic cells, but not those from painful discs, was reported. The mechanical and biological function of these processes remains unknown.

This study is important for revealing that cell responses to dynamic loading, particularly cell death and proteoglycan content, are dependent on both the frequency and magnitude of the applied stress. The lower frequency and higher peak stress magnitudes induced the greatest increases in proteoglycan content and cell death that may relate to the higher deformations that occur under these conditions.

When the mechanically compressed nucleus pulposus tissue was transplanted to the rat nerve roots, the animals experienced a decreased sensitization to pain, providing some of the first evidence that mechanical stimuli have the potential to regulate the perception of pain.

    • Cyclic tensile stretch modulates proteoglycan production by intervertebral disc annulus fibrosus cells through production of nitrite oxide
    • Rannou F, Richette P, Benallaoua M, et al.: Cyclic tensile stretch modulates proteoglycan production by intervertebral disc annulus fibrosus cells through production of nitrite oxide. J Cell Biochem 2003, 90:148-157. This study is one of the first to study a mechanism for the inhibition of proteoglycan production in IVD cells by tensile stretch. Using a set of nitric oxide synthase inhibitors, the authors conclude that a specific form of nitric oxide synthase, cNOS, may be involved in the downregulation of proteoglycan synthesis, and upregulation of nitric oxide, following tensile stretch.
