This important article presented the first evidence that homozygous mutations in the newly recognized hemojuvelin gene are the cause of most cases of juvenile hemochromatosis, with the remainder due to homozygous mutations in the hepcidin gene. Patients with juvenile hemochromatosis due to hemojuvelin mutations have very low hepcidin excretion suggesting that hemojuvelin normally acts as a regulator of hepcidin.
