Co-localization of MTOC and Gag were demonstrated using indirect immunofluorescence in infected cells and EM analysis. A direct interaction between Gag and LC8 constitutes one of the molecular bases of such trafficking.

The transduction was demonstrated by long-term transgene expression in first and second transplant-recipient mice without gene silencing, demonstrating that FV-derived vectors might be a safe and effective alternative to MLV or lentivirus vectors in human gene therapy.

This is the first direct comparison of vectors of the three classes of retroviruses. It illustrates that both the cellular activation state and the progression through the cell cycle regulate the ability of retroviral vectors to efficiently deliver the foreign transgenes. Finally, they suggest that FV vectors might be the best choice for ex vivo stem cell gene therapy because they are equal to HIV vectors, but without safety concerns, and superior to MLV vectors in their ability to transduce transiently quiescent cells.
