Gilmore TD, Herscovitch M. Inhibitors of NF-κB signaling: 785 and counting. Oncogene 2006; 25:6887-6899. This article provides an excellent review on 785 NF-κB inhibitors which have been used to target upstream or downstream NF-κB signaling in cultured cells or tissues. The information is particularly useful to investigators for selection of NF-κB inhibitors to block different steps of NF-κB activation in their studies.

Mol Endocrinol 2007; 21:2203-2217. This paper focuses on the role of the Rac/RhoA GTPase-NF-κB inducing kinase (NIK) signaling pathway in mediating Ang II-induced RelA activation, independent of the classical IκB degradation, in hepatocytes.

McAllister-Lucas LM, Ruland J, Siu K, et al. CARMA3/Bcl10/MALT1- dependent NF-κB activation mediates angiotensin II-responsive inflammatory signaling in nonimmune cells. Proc Natl Acad Sci U S A 2007; 104:139-144. This paper characterized a previously undescribed signaling pathway mediating Ang II-dependent activation of NF-κB signaling. This pathway is composed of three principal proteins: CARMA3, Bcl10 and MALT1. Inhibition of these proteins with dominant negative mutants, siRNA or gene targeting effectively blocked Ang II-induced NF-κB activation in hepatocytes.

Henke N, Schmidt-Ullrich R, Dechend R, et al. Vascular endothelial cellspecific NF-κB suppression attenuates hypertension-induced renal damage. Circ Res 2007; 101:000-10. The authors generated Cre/lox transgenic mice with endothelial cell-restricted NF-κB super-repressor IκBαΔN (Tie-1-ΔN mice). They were able to show for the first time that vascular endothelial cell-specific NF-κB suppression can attenuate L-NAME- or Ang II-induced hypertensive renal damage.
