Mouse limb deformity mutations disrupt a global control region within the large regulatory landscape required for Gremlin expression

Several Id alleles are used in combination with Grem1 and Fmn1 loss-of-function mutations (generated by gene targeting) to establish that the recessive mouse Id phenotype is caused by disrupting a cis-regulatory global control region embedded in the Fmn1 gene. The authors map the critical region by BAC transgenesis and show that the cis-regulatory elements in Fmn1 are required for limb specific expression of Grem1, which in turn is essential for distal limb development. This study demonstrates how difficult the correct assignment of a mutant phenotype can be if it is caused by disruption of an essential cis-regulatory element inserted in an unrelated other gene.

A series of ENU-induced single-base substitutions in a long-range cis-element altering Sonic hedgehog expression in the developing mouse limb bud

Using a genomic DNA archive of ENU-mutagenized mice, the authors have identified novel point mutations in the MFCS1 region. Several of the point mutations result in PPD with associated anterior Shh ectopic expression while others are silent and do not affect Shh expression. Transgenic mouse embryos expressing reporters under control of the functionally altered mutant MFCS1 sequences show that they retain their gain-of-function phenotype even out of their normal genomic context. Screening for the essential sequences within the MFCS1 using, for example gene targeting would be tedious, while the ENU approach used in this study seems rather effective. Therefore, the existing ENU DNA archive is a useful resource for the analysis of other genomic regions of interest.

Using specific antibodies against Hoxd10 and Hoxd13 proteins for ChIP analysis of limb buds, the authors demonstrate that they are part of a complex bound to a core ZRS/MFCS1 DNA fragment (called ShhE). Furthermore, Hoxd proteins are able to activate transcription of a ShhE-reporter construct in transfected cells. These important results are in line with the fact that genetic analysis has indicated that the ZRS/MFCS1 functions to activate and positively regulate Shh expression in limb buds.
