Pascual G, Fong AL, Ogawa S, et al. A SUMOylation-dependent pathway mediates transrepression of inflammatory response genes by PPAR-gamma. Nature 2005; 437:759-763. This study is a very interesting and important report in terms of receptor protein modifications and corepressor recruitment. Ligand binding to peroxisome proliferator-activated receptor γ causes a structural change. This results in the exposure of a previously hidden region in the ligand binding domain. When this region is SUMOylated, protein modification by SUMO modifies the receptor's ability to stably associate with corepressors. This is the first description of the physiological significance of SUMOylation of receptors - a modification proven to be associated with ligand-induced transcriptional repression.
