A high-density screen for linkage in multiple sclerosis
-
SNP-based typing, in the largest linkage analysis done to date, has failed to show any significant loci in non-HLA gene regions, suggesting low odds ratios of regulating genes, and that future genetics in MS will largely depend on association analysis in very large cohorts, either based on candidate genes or whole genome.

Two genes encoding immune-regulatory molecules (LAG3 and IL7R) confer susceptibility to multiple sclerosis

Around 70 candidate genes where chosen for SNP typing on the basis of the presence of suggestive linkage peaks and/or immunological function in a fairly large case-control material. MHC2TA is associated with differential MHC molecule expression and susceptibility to rheumatoid arthritis, multiple sclerosis and myocardial infarction
-
By positional cloning of Vra4, a QTL on rat chromosome 10 that regulates MHC class II expression after nerve injury, the authors have identified the regulatory gene, MHC class II transactivator (Mhc2ta). The polymorphism in the type III promoter of Mhc2ta confers susceptibility to several common complex diseases with inflammatory components, including multiple sclerosis, rheumatoid arthritis and myocardial infarction.
