Depletion of the human homolog of Dasra B (Borealin/Dasra B [17]) resulted in severe chromosome alignment defects in HeLa cells. • Add-back of several INCENP mutants in INCENP-depleted cells demonstrated that the first 47 amino acids of INCENP bind Survivin and that a fusion protein of Survivin and INCENP, lacking the first 47 amino acids, is sufficient to target a functional passenger complex to the centromere and central spindle in the absence of endogenous INCENP and Borealin. • Chromosome alignment and segregation regulated by ubiquitination of survivin
    • In a Survivin immunoprecipitate from X. laevis extracts both a deubiquitinating enzyme (FAM) and a complex (p97-NP14-Ufd1) involved in ubiquitination were found as novel interaction partners of Survivin. Survivin is ubiquitinated in mitosis and this appears to regulate its dynamic association with the centromere. Ubiquitination of Survivin by the p97-NP14-Ufd1 complex is needed for association with the centromere, whereas its deubiquitination by FAM is required for its dissociation. This dynamic behavior of Survivin appears to be important for its centromere function.

    • This paper shows that the effect of BubR1 depletion on chromosome-spindle attachments, as originally shown by Ditchfield et al. [21], can be restored by inhibition of Aurora B kinase activity. • Phosphorylation of ZEN-4/MKLP1 by aurora B regulates completion of cytokinesis
    • The kinesin-6 protein MKLP1/ZEN-4 is part of the central spindlin complex essential for central spindle assembly. This paper demonstrates that MKPL1 is a substrate of Aurora B and that expression of a non-phosphorylatable mutant of ZEN-4 in C. elegans embryos affects the later stages (completion) of cytokinesis. In mammalian cells, inhibition of Aurora B kinase inhibited MKLP1 phosphorylation in vivo. Addition of the kinase inhibitor to anaphase cells did not perturb the early events of cytokinesis (furrow ingression), but eventually furrows regressed. Furthermore, even when Aurora B was inhibited during telophase, when the midbody was formed, it inhibited the completion of cytokinesis.

    • Centromere targeting of the chromosomal passenger complex requires a ternary subcomplex of borealin, survivin, and the N-terminal domain of INCENP
    • This paper demonstrates that a 58 amino acid N-terminal fragment of INCENP interacts with both Survivin and Borealin and that Borealin can interact with double-stranded DNA in vitro. It is suggested that a complex of Survivin, Borealin and the N-terminus of INCENP is necessary for centromere targeting of the CPC.
