This study used mast cell knock-in mice and other lines of evidence to show that mast cell-derived IL-4 can promote Th1 cell development in a mouse model of experimental allergic encephalomyelitis. The results presented suggest that the site and timing of IL-4 expression, as well as the genetic background of the mice used, can contribute to the heterogeneity of the T helper response elicited in this model of autoimmunity.

This study employed mast cell knock-in mice to demonstrate that mice that had been bitten by mosquitoes before subcutaneous sensitization to OVA (at another skin site) exhibited a mast cell-dependent suppression of antigen-specific DTH responses to OVA when they were later challenged with OVA at the same site as the mosquito bite. The mast cell-dependent immunoregulatory mechanism in this model remains to be defined, including identification of the extent to which key effects occur in the sensitization versus the elicitation phase of the DTH response. However, the authors detected enhanced levels of IL-10 mRNA in draining LNs 24 h after the mosquito bite. Moreover, administration of anti-IL-10 antibody before sensitization with OVA restored the magnitude of DTH responses, as well as levels of skin and LN IFN-γ mRNA and protein, in mosquito-bitten mice to values similar to those observed in control unbitten mice.

This study showed that, in mice bearing antigen-specific IgE that were challenged with a single subcutaneous injection of the specific multivalent antigen, mast cells were essential for IgE-dependent immediate and late phase ear-swelling responses. However, it showed that they were not essential for the delayed-onset ear-swelling response that was characterized by persistent inflammation and extensive eosinophil infiltration. Transfer of FcεRI-expressing basophils into FcεRI-deficient mice restored the ability to develop the delayed-onset phase of such allergic inflammation. This finding, together with other lines of evidence in this study, supports the conclusion that in this model basophils played an important role, independent of T cells and mast cells, in the development of IgE-dependent chronic allergic inflammation.
