Cellular and genetic mechanisms of self tolerance and autoimmunity
-
Goodnow CC, Sprent J, Fazekas de St Groth B, Vinuesa CG. Cellular and genetic mechanisms of self tolerance and autoimmunity. Nature 2005; 435:590-597. This paper highlights mechanisms, including those on a genetic level, involved in self-tolerance. Similar mechanisms may presumably occur following in-utero transplantation for tolerance induction.

+ T cells (Tc2 cells) is not sufficient to improve engraftment in fetal transplantation. Fetal Diagn Ther 2005; 20:35-43. This paper evaluates a cell population with the potential to increase the incidence of in-utero tolerance induction. The cell population has been shown in postnatal studies to have an immune suppressive function. Therefore, this paper highlights, among other things, a potential fetal immune barrier to tolerance induction.

Long-term acceptance of renal allografts following prenatal inoculation with adult bone marrow
-
Mathes DW, Solari MG, Randolph MA, et al. Long-term acceptance of renal allografts following prenatal inoculation with adult bone marrow. Transplantation 2005; 80:1300-1308. This paper describes one of the few studies demonstrating, in a large animal model, that in-utero tolerance induction can serve as a platform for postnatal organ transplantation. The technique used for in-utero injection (injection through the intrahepatic portion of the umbilical vein) and the composition of T cells in the donor cell inoculum are different from those of other groups published and may contribute to their excellent results.

Busulfan-conditioned bone marrow transplantation results in high-level allogeneic chimerism in mice made tolerant by in utero hematopoietic cell transplantation

Busulfan-conditioned bone marrow transplantation results in high-level allogeneic chimerism in mice made tolerant by in utero hematopoietic cell transplantation. Exp Hematol 2006; 34:359-368. This paper highlights the ability of in-utero tolerance induction to allow for successful nontoxic postnatal allogeneic cellular transplantations. It highlights the correlation between postnatal chimerism levels and tolerance induction following in-utero transplantation.

Muench MO. In utero transplantation: baby steps towards an effective therapy. Bone Marrow Transplant 2005; 35:537-547. -
Rizzia M, Gerloni M, Srivastava AS, et al. In utero DNA immunisation. Immunity over tolerance in fetal life. Vaccine 2005; 23:4273-4282. This study highlights that it is possible to immunize the fetus, despite the previous held belief that the immaturity of the developing fetal immune system prevents it from such a process. This study is important to the field of in-utero transplantation and tolerance induction because it forces one to consider the fetal immune system as a potential barrier to engraftment.

Stable multilineage chimerism across full MHC barriers without graft-versus-host disease following in utero bone marrow transplantation in pigs
-
Lee PW, Cina RA, Randolph MA, et al. Stable multilineage chimerism across full MHC barriers without graft-versus-host disease following in utero bone marrow transplantation in pigs. Exp Hematol 2005; 33:371-379. This study evaluated the role of T cells in the donor innoculum for in-utero transplantation in a large animal model. It is of great interest to those whose goal it is to translate in-utero tolerance induction more successfully into the clinics, as their techniques for donor-cell processing and composition have yielded the most consistent successful results to date.

B72-/- mature dendritic cells generate T-helper 2 and regulatory T donor cells in fetal mice after in utero allogeneic bone marrow transplantation
-
Bhattacharyya S, Cowan MJ. B72-/- mature dendritic cells generate T-helper 2 and regulatory T donor cells in fetal mice after in utero allogeneic bone marrow transplantation. Biol Blood Marrow Transplant 2005; 11:657-671. The addition of a facilitating cell population to the donor-cell inoculum is one way to potentially overcome barriers to in-utero tolerance induction. This article is of importance to those interested in the study of in-utero tolerance induction as it highlights this possibility and mandates that the reader evaluate other cell populations.

+ mobilized peripheral blood as a donor cell source does not improve chimerism after in utero hematopoietic stem cell transplantation in nonhuman primates. J Med Primatol 2005; 34:201-208. This study is of interest to those studying in-utero tolerance induction because it highlights the inability of the donor cell to compete with the host cell as a barrier to engraftment and forces one to evaluate the benefits of different donor-cell populations.

Hematopoietic microchimerism in sheep after in utero transplantation of cultured cynomolgus embryonic stem cells
-
Sasaki K, Nagao Y, Kitano Y, et al. Hematopoietic microchimerism in sheep after in utero transplantation of cultured cynomolgus embryonic stem cells. Transplantation 2005; 79:32-37. This study is of great interest to those studying in-utero tolerance induction because it highlights the potential of the fetal immune system to serve as a barrier to engraftment and provides evidence that it is possible to sensitize, not tolerize, the fetus after in-utero transplantation.
