Positive association of DTNBP1 with schizophrenia in a large case-control sample collected from the US is described in this study. The samples consisted of a subset of 258 Caucasian patients and 467 controls and smaller subsets of African Americans and Hispanics. This paper provides an excellent summary of the different haplotypic associations observed in the DTNBP1 literature.

Hum Mol Genet 2005; 14:1947-1954. This study uses allelic expression assays to examine DTNBP1 mRNA levels. In concordance with Weickert et al. [17], the authors found that reduced DTNBP1 expression is associated with the same polymorphism in the 30-UTR. This study also shows that a defined schizophrenia risk haplotype in DTNBP1 and risk haplotypes identified in other sample groups tag cis-acting variants that results in a relative reduction of DTNBP1 mRNA.

Am J Med Genet B Neuropsychiatr Genet 2005 [Epub ahead of print]. The authors of this study genotyped 11 SNPs across the DAOA (G72) locus in a sample of 233 Palestinian Arab families. The same markers were previously genotyped by this group in a small sample of Ashkenazi Jews. Both studies showed significant association with schizophrenia, but in Palestinian Arabs, significant association was observed with a single SNP, with ∼8 kb from the associated linkage disequilibrium block in the Ashkenazi sample and with an adjacent haplotype.

This is the most recent meta-analysis of the COMT Val158Met polymorphism with schizophrenia in case-control samples. The analyses encompass data from 18 studies, nine of which are of European ancestry. The analyses do not support association of schizophrenia with Val158Met.

Following the reports of strong haplotypic association of COMT in Ashkenazi Jewish and Irish subjects, the authors of this study attempted replication. In both, a UK-based case-control sample of 709 schizophrenic patients and 710 controls and a second independent sample of 488 parent-proband trios from Bulgaria, the authors found no evidence for association of schizophrenia with the COMT gene with any single marker or haplotype and no association with gender.

This study includes data from eight markers spanning not only the COMT gene but also the adjacent genes, TR2 and ARVCF. Analyses in 136 schizophrenia families showed association with a three-marker haplotype spanning COMT and ARVCF suggesting that ARVCF may also contribute to schizophrenia susceptibility.

Following the initial report of association of ZDHHC8 with schizophrenia [56], the authors of this study performed association analyses with rs175174 in 465 schizophrenia patients and 467 controls and a 158 parent-proband trios of Han Chinese origin. The authors found significant association with rs175174 allele G in both samples, contrary to the original positive study [56].

In this study, 407 schizophrenia patients and 497 controls of Japanese origin showed no evidence for association of rs175174 in ZDHHC8 with schizophrenia or the presence of gender-specific differences. The same analyses were also negative with bipolar disorder and when the sample of schizophrenia patients was classified into paranoid and disorganized types.

In this paper, analyses of a Caucasian case-control sample of 196 schizophrenic, 82 bipolar, 62 schizoaffective patients and 217 controls from North America showed that a common haplotype is associated with schizoaffective disorder. This study supports the hypothesis that the DISC1 locus predisposes to a variety of psychiatric disorders and that schizophrenia and bipolar disorder may have convergent aetiology.
