Using a strategy of disruption of disulphide bonds, a hypoallergenic recombinant Hev b 6.01 mutant was produced. A synthetic peptide based on the cysteine-substitituted Hev b 6.02 domain of this mutant lacked IgE binding as tested by inhibition ELISA and basophil activation assay but retained T-cell reactivity, consistent with a candidate hypoallergenic preparation.
