Reactive oxygen species from smooth muscle mitochondria initiate cold-induced constriction of cutaneous arteries
-
Epub ahead of print
-
Bailey SR, Mitra S, Flavahan S, et al. Reactive oxygen species from smooth muscle mitochondria initiate cold-induced constriction of cutaneous arteries. Am J Physiol Heart Circ Physiol 2005 [Epub ahead of print]. This extremely important study confirms that the initial trigger for Rho/Rho kinase signaling is provided by a rapid increase of ROS in smooth muscle cells after cold exposure. ROS inhibitors were able to abolish cold-induced increase in α2c-adrenoreceptor constrictor activity. These results prompt reconsideration of anti-oxidant treatment strategies in Raynaud's phenomenon.

Distinct cAMP signaling pathways differentially regulate alpha2C-adrenoceptor expression: Role in serum induction in human arteriolar smooth muscle cells

Am J Physiol Heart Circ Physiol 2005; 288:H69-H76. Findings in this study suggest that exposure to serum induces expression of α2c-adrenergic receptors on VSMs. Similarly enhanced vasoconstrictive responses to cooling can result in vivo from traumatic or inflammatory events exposing VSMs to serum.

Digital ulcers in systemic sclerosis: Prevention by treatment with bosentan, an oral endothelin receptor antagonist
-
Korn JH, Mayes M, Matucci Cerinic M, et al. Digital ulcers in systemic sclerosis: prevention by treatment with bosentan, an oral endothelin receptor antagonist. Arthritis Rheum 2004; 50:3985-3993. This study provides evidence that endothelin-1 inhibitors can prevent ischemic ulcers in scleroderma. No clinical benefit was shown in terms of number and severity of Raynaud's phenomenon attacks, however.
