Polymorphisms in the Fc epsilon RI beta promoter region affecting transcription activity: A possible promoter-dependent mechanism for association between Fc epsilon RI beta and atopy
-
C. Nishiyama, Y. Akizawa, M. Nishiyama, T. Tokura, H. Kawada, K. Mitsuishi, M. Hasegawa, T. Ito, N. Nakano, and A. Okamoto Polymorphisms in the Fc epsilon RI beta promoter region affecting transcription activity: a possible promoter-dependent mechanism for association between Fc epsilon RI beta and atopy J Immunol 173 2004 6458 6464 The controversy involving a possible SNP in the coding region of the FcRβ gene, which was previously linked to allergic symptoms and FcεRI expression level, and the finding that the resulting mutation does not by itself modulate this expression, is solved in this work. Indeed, the authors describe additional SNPs in the FcRβ gene promoter region, linked to the previously described SNP, leading to enhanced promoter activity and thereby enhanced FcεRI expression in atopic individuals.

Negative regulation of immunoglobulin E-dependent allergic responses by Lyn kinase
-
S. Odom, G. Gomez, M. Kovarova, Y. Furumoto, J.J. Ryan, H.V. Wright, C. Gonzalez-Espinosa, M.L. Hibbs, K.W. Harder, and J. Rivera Negative regulation of immunoglobulin E-dependent allergic responses by Lyn kinase J Exp Med 199 2004 1491 1502 This work dissects the mechanisms responsible for the allergic-like phenotype of Lyn-deficient mice. The study shows how this src kinase phosphorylates an adaptor that recruits a kinase which inactivates src kinases by phosphorylating a regulatory tyrosine of these enzymes. It also shows how, by reducing IL-4 signalling in B cells, Lyn prevents high titers of IgE from being synthesized and, as a consequence, controls mast cell numbers and FcεRI expression.

Identification of FcalphaRI as an inhibitory receptor that controls inflammation: Dual role of FcRgamma ITAM
-
B. Pasquier, P. Launay, Y. Kanamaru, I.C. Moura, S. Pfirsch, C. Ruffie, D. Henin, M. Benhamou, M. Pretolani, and U. Blank Identification of FcalphaRI as an inhibitory receptor that controls inflammation: dual role of FcRgamma ITAM Immunity 22 2005 31 42 An ITAM-bearing receptor that activates cells upon aggregation by multivalent ligands was found to inhibit cell activation induced by other receptors upon ligation by monovalent ligands. Monovalent binding induced a weak phosphorylation of ITAMs, which recruited the tyrosine phosphatase SHP-1. The significance of this finding will increase if it is reproduced in more physiological models.
