Sykulev Major histocompatibility complex class I-intercellular adhesion molecule-1 association on the surface of target cells: implications for antigen presentation to cytotoxic T lymphocytes Immunology 113 2004 460 471 In this paper, the authors have shown that productive engagement of ICAM-1 on target cells results in concomitant recruitment of rafts and MHC class I molecules (e.g. HLA-A2) to the interface between the target cells and surrogate CTLs (beads loaded with anti-ICAM-1 antibodies). In addition, disruption of raft integrity on 'live' target cells leads to a release of ICAM-1 and HLA-A2 from the raft fraction and impairs the ability of target cells to present viral peptides to CTLs. The data suggest that ICAM-1 co-stimulates peptide presentation to CTLs.

As the IS matures, relevant pMHC-II complexes are sorted to a central region, whereas irrelevant pMHC-II molecules are excluded from this area. The authors hypothesize that, at limited peptide density, irrelevant pMHCs are involved in the initial stabilization of APC-T cell contact.

    • J. Greenwood, C.L. Amos, C.E. Walters, P.O. Couraud, R. Lyck, B. Engelhardt, and P. Adamson Intracellular domain of brain endothelial intercellular adhesion molecule-1 is essential for T lymphocyte-mediated signaling and migration J Immunol 171 2003 2099 2108 By transfecting brain endothelial cells with either wild-type ICAM-1 or ICAM-1 with defective intracellular domain, the authors have shown that the effectiveness of transendothelial migration of T lymphocytes, but not their adhesion to the endothelial cells, was dependent on ICAM-1-mediated signaling leading to Rho GTPase activation. The data indicate that, in addition to adhesion, ICAM-1 induces a signal that facilitates migration of T lymphocytes across the endothelial barrier.
