In addition, dominant-negative mutant Vps4 proteins that block vacuolar protein sorting also arrest HIV-1 budding. Their results provide a proof-of-concept in that Tsg101/Vps proteins are involved in HIV assembly/budding.

The researchers used proteasome inhibitors to treat cells transfected with an HIV-1 proviral DNA construct. This treatment reduced intracellular levels of free ubiquitin by trapping conjugated ubiquitin inside the blocked proteasomes, preventing it from being recycled. The reduction in ubiquitin content resulted in a block to HIV-1 particle release and a significant reduction in viral infectivity.
