This study indicates that the metabolic pathway most sensitive to cyclosporine is high-energy phosphate metabolism, and that sirolimus synergistically enhances these negative effects of cyclosporine.

J Cereb Blood Flow Metab (2002) 22(3):342-352. This study demonstrates that the neuroprotective or neurotoxic nature of the effects of cyclosporine depends on both the cyclosporine and oxygen concentrations. The results also imply that cyclosporine-induced neurotoxicity and neuroprotection during hypoxia/reperfusion may both be caused by inhibition of calcium traffic through the mitochondrial membrane.

Br J Pharmacol (2001) 133(6):875-878. This study demonstrates that cyclosporine inhibits the Krebs cycle and mitochondrial oxidative phosphorylation in vivo. While co-administration of sirolimus enhances the negative effects of cyclosporine, co-administration of the same dose of everolimus antagonizes them.

J Pharmacol Exp Ther (1996) 277(1):417-422. This study demonstrates that application of cyclosporine to rat liver mitochondria in contact with calcium induced both an accumulation of calcium and a decrease in oxidative phosphorylation. The interaction of cyclosporine with mitochondrial calcium fluxes is probably the first event that triggers all the metabolic changes described in this review.

This study demonstrates that treatment of Wistar rats with cyclosporine (10 mg/kg/day for 6 days) negatively affected mitochondrial metabolism in all organs, although the metabolic responses were different, with the kidney being one of the most sensitive organs and the liver being the least sensitive.
