In situ hybridization of the α6 and β3 subunits in rat brain demonstrated that these subunits are expressed at high levels in catecholaminergic neurons. Semi-quantitative analysis showed that the mRNA coding for these subunits are actually more abundant than the mRNA coding for all other subunits expressed in the structure. These results suggest that α6β3β2-containing nAChRs contribute to the nicotinic responses of dopaminergic neurons.

This key work studies the effect of a mutation in the channel domain of the nAChR ε subunit previously identified by the authors as being responsible for a human congenital myasthenic syndrome. The transgenic mice exhibit a wide spectrum of neurophysiological and histological defects highly reminiscent of, if not identical to, the defects observed in humans. This provides a clear demonstration of the cytotoxicity of a mutation increasing the open-channel duration of muscle nAChRs.

This mutation results from the insertion of a leucine in the transmembrane domain lining the ion channel. This paper shows that β2-containing nAChRs increase the spontaneous and evoked release of GABA in the mouse thalamus. This effect is attributable to an influx of calcium in the nerve terminal. Different pathways of calcium influx are recruited, depending on the type of nerve terminal.
