This paper provides the first indication that the α6 nicotinic subunit may contribute to a functional nicotinic receptor. In situ hybridization of the α6 and β3 subunits in rat brain demonstrated that these subunits are expressed at high levels in catecholaminergic neurons. Semi-quantitative analysis showed that the mRNA coding for these subunits are actually more abundant than the mRNA coding for all other subunits expressed in the structure. These results suggest that α6β3β2-containing nAChRs contribute to the nicotinic responses of dopaminergic neurons.

This key work studies the effect of a mutation in the channel domain of the nAChR ε subunit previously identified by the authors as being responsible for a human congenital myasthenic syndrome. The transgenic mice exhibit a wide spectrum of neurophysiological and histological defects highly reminiscent of, if not identical to, the defects observed in humans. This mutation results from the insertion of a leucine in the transmembrane domain lining the ion channel. The effect of this mutation on the function of the receptor is examined in oocyte experiments.

Nature 1996, 382:255-257. 2-deoxyglucose autoradiography and brain microdialysis demonstrate a specific increase of metabolism and release of dopamine in the shell of the nucleus accumbens following injection of minimal doses of nicotine. Similar effects have also been described for cocaine, amphetamine and morphine, and are likely to be associated with the addictive properties of these drugs. These results, though not acquired in the context of nicotine self-administration, are thus strongly in favour of an addictive action of nicotine.

Regional brain activation following nicotine and cocaine self-administration is assessed by mapping Fos-like immunoreactivity. Similar patterns of activation are observed for both drugs in the terminal fields of the mesocorticolimbic system, supporting the view that self-administration of nicotine is associated with an activation of the mesencephalic dopaminergic system, as observed for other reinforcing drugs.
