Structure of dengue virus: Implications for flavivirus organization, maturation, and fusion

Cryo-EM reconstruction of the dengue flavivirus. The glycoprotein E crystal structure from closely related tick-borne encephalitis virus was fitted into the EM density to produce a pseudo-atomic model. Although 180 copies of the E protein are present in the outer icosahedral shell, they do not conform to the expected T=3 quasi-equivalent organization. Instead, 90 dimers of the E protein are arranged in a 'herringbone' pattern on the surface. The inner nucleoprotein core contained within a lipid bilayer is apparently somewhat variable in orientation or less ordered. Based upon the pseudo-atomic model of the outer shell, membrane fusion mechanisms are discussed

The virus was also titrated as a function of pH and the transition was found to occur over a narrow 0.7 pH unit window
