Cholesteryl ester transfer protein expression attenuates atherosclerosis in ovariectomized mice
-

Cazita PM, Berti JA, Aoki C, Gidlund M, Harada LM, Nunes VS, Quintao ECR, Oliveira HCF: Cholesteryl ester transfer protein expression attenuates atherosclerosis in ovariectomized mice. J Lipid Res (2003) 44(1):33-40. This study addressed the possibility that the CETP gene and estrogen interact. CETP deficiency increased atherosclerosis in ovariectomized mice, which was mediated by increased CE uptake and decreased cholesterol efflux activity in macrophages, as well as increased plasma levels of atherogenic lipoproteins.

Cholesteryl ester transfer protein inhibitor (JTT-705) and the development of atherosclerosis in rabbits with severe hypercholesterolaemia

note

Huang Z, Inazu A, Nohara A, Higashikata T, Mabuchi H: Cholesteryl ester transfer protein inhibitor (JTT-705) and the development of atherosclerosis in rabbits with severe hypercholesterolaemia. Clin Sci (2002) 103(6):587-594. CETP inhibition did not reduce atherosclerosis in cholesterol-fed rabbits in which non-HDL cholesterol was inadequately decreased. This study indicates that more research is needed on the mechanisms of TG metabolism and post-prandial hyperlipidemia when CETP is inhibited.

Efficacy and safety of a novel cholesteryl ester transfer protein inhibitor, JTT-705, in humans: A randomized phase II dose-response study
-
note
-
De Grooth GJ, Kuivenhoven JA, Stalenhoef AFH, De Graaf J, Zwinderman AH, Posma JL, Va Tol A, Kastelein JJP: Efficacy and safety of a novel cholesteryl ester transfer protein inhibitor, JTT-705, in humans: A randomized phase II dose-response study. Circulation (2000) 105(18):2159-2165. This is the first clinical trial on JTT-705 reported. JTT-705 clearly increased HDL-C levels and decreased plasma CETP activity. The drug increase HDL3 as well as HDL2. A mild reduction of LDL was observed, especially in individuals with baseline LDL-C > 170 mg/dl.

Apolipoprotein E mediates the retention of high-density lipoproteins by mouse carotid arteries and cultured arterial smooth muscle cell extracellular matrices
-
note
-
Olin-Lewis K, Benton JL, Rutledge JC, Baskin DG, Wight TN, Chait A: Apolipoprotein E mediates the retention of high-density lipoproteins by mouse carotid arteries and cultured arterial smooth muscle cell extracellular matrices. Circ Res (2002) 90(12):1333-1339. HDL together with apoE, which is increased in CETP deficiency, was trapped in the vascular extracellular matrix, indicating that HDL with apoE has direct effects on the artery wall. However, whether these particles are anti- or pro-atherogenic, is not yet known.
